Jonathan Spence studerte først historie ved [[UniversitetetUniversity iof Cambridge]]. Sinologien kom han i berøring med under et utvekslingsår ved [[Yale University]], der forelesninger av historieprofessor Mary Wright om kinesisk historie vekket hans interesse. Han tok doktorgraden med en avhandling om den tidlige [[Qing-dynastiet|Qing-tiden]], og avhandlingen ble fulgt opp med verket ''To Change China: Western Advisers in China''. Etter dette fikk han i 1971 en lærestol i historie ved Yale. Flere publikasjoner fulgte.

Mellom 1983 og 1986 var Spence dekan for detYales historiske fakultet, og i årene 1988/891989 ledet han [[Whitney Humanities Center]].
